What the Avian Influenza Virus H9 kit detects


The AICON-BIO® Avian Influenza Virus H9 Real-time PCR Kit uses real-time PCR technology and a premixed freeze-drying method to detect avian influenza virus subtype H9 nucleic acid. H9N2 is the most widely distributed low-pathogenic avian influenza subtype in commercial poultry across Asia, the Middle East and North Africa, where it is now effectively endemic.

Although classed as low-pathogenic, H9N2 causes heavy losses in combination with secondary bacterial infection, and it has repeatedly donated internal gene segments to H5 and H7 reassortants — as well as infecting humans directly. Routine H9 monitoring is therefore a One Health priority, not only a production one. The Kit qPCR mix (lyophilized) contains primers, probes, enzyme and dNTPs in one tube.

Clinical context. H9N2 — endemic LPAI, gene donor to H5/H7.

Workflow

Four steps from extract to result


  1. Extract nucleic acid with any commercial extraction kit.
  2. Reconstitute one lyophilized tube with the supplied buffer, mix, spin briefly.
  3. Combine 20 µL reconstituted reagent + 5 µL template (or control) = 25 µL reaction.
  4. Run the FAM-channel cycling programme; the instrument software reads the result.

Set the reaction volume to 25 µL and select the FAM channel. On ABI systems leave the
analysis threshold at its default value of 0.2.

Run validity. The negative control must show

Specimen collection & storage


TissueGreen-bean-sized piece into a 1.5 mL tube with 500 µL preservation solution; grind with a disposable pestle; centrifuge 1 min; take the supernatant.
Whole bloodCollect into an EDTA anticoagulant tube.
SerumAllow the blood to clot or centrifuge at low speed; collect the supernatant.
SwabSmear the sampling site, place the tip into a 1.5 mL tube with 500 µL preservation solution; grind a few seconds; centrifuge 1 min; take the supernatant.

Storage & stability

Lyophilized reagents ship at room temperature — no cold chain required. On arrival, store the lyophilized reagents at −20 °C; liquid reagents below 4 °C. Stable until the expiry date printed on the label. Keep the tubes sealed in their aluminium pouches to prevent moisture uptake and bring them to room temperature before use. Once reconstituted, seal tightly, store at −20 °C and use within one month.
