What the Avian Influenza Virus H5 kit detects
The AICON-BIO® Avian Influenza Virus H5 Real-time PCR Kit uses real-time PCR technology and a premixed freeze-drying method to detect avian influenza virus subtype H5 nucleic acid. Highly pathogenic H5 viruses of the goose/Guangdong lineage — H5N1 clade 2.3.4.4b in particular — have spread worldwide through wild birds, causing mass mortality in poultry and wild populations and spilling over into an unprecedented range of mammals.
H5 infection is WOAH-notifiable and closes export markets on confirmation, so laboratories need a subtype-specific assay that returns an answer in hours. The Kit qPCR mix (lyophilized) contains primers, probes, enzyme and dNTPs in one tube.

Workflow
Four steps from extract to result
- Extract nucleic acid with any commercial extraction kit.
- Reconstitute one lyophilized tube with the supplied buffer, mix, spin briefly.
- Combine 20 µL reconstituted reagent + 5 µL template (or control) = 25 µL reaction.
- Run the FAM-channel cycling programme; the instrument software reads the result.

Set the reaction volume to 25 µL and select the FAM channel. On ABI systems leave the
analysis threshold at its default value of 0.2.
Result interpretation
Reading the run

no amplification and the positive control a clear amplification curve. If the negative control

Specimen collection & storage
| Tissue | Green-bean-sized piece into a 1.5 mL tube with 500 µL preservation solution; grind with a disposable pestle; centrifuge 1 min; take the supernatant. |

Storage & stability
Lyophilized reagents ship at room temperature — no cold chain required. On arrival, store the lyophilized reagents at −20 °C; liquid reagents below 4 °C. Stable until the expiry date printed on the label. Keep the tubes sealed in their aluminium pouches to prevent moisture uptake and bring them to room temperature before use. Once reconstituted, seal tightly, store at −20 °C and use within one month.
